Tacgraf 1mg Capsule 10 ‘S is an immunosuppressant agent. It is used with other medicines in people who have had a heart, kidney, liver, or lung transplant. Tacgraf 1mg Capsule 10 ‘S weakens your immune system to prevent the body from rejecting the transplanted organ. Tacgraf 1mg Capsule 10 ‘S comes in capsule form. 

Side effects such as headache, diarrhea, nausea, vomiting, and upset stomach may occur after the use of Tacgraf 1mg Capsule 10 ‘S. Before taking Tacgraf 1mg Capsule 10 ‘S tell your doctor if you are allergic to the active substance of Tacgraf 1mg Capsule 10 ‘S or to other macrolide drugs. 

Tacgraf 1mg Capsule 10 ‘S works by inhibiting the activity of T-lymphocytes. Tacgraf 1mg Capsule 10 ‘S is taken orally after transplant surgery has taken place. The Tacgraf 1mg Capsule 10 ‘S is given in allogeneic transplants. This is a procedure in which a patient receives healthy blood-forming cells (stem cells) from a donor to replace their stem cells that have been destroyed by cancer treatment. The recipient’s cells are destroyed with radiation or high doses of chemotherapy (cancer drugs). 

Indications

Tacgraf 1mg Capsule 10 ‘S can be used for:

  • Prophylaxis of graft rejection, in the allogeneic liver, kidney, or heart transplantation.

How Does Tacgraf 1mg Capsule 10 ‘S Work

{{product’s}} mechanism of action is based on calcineurin inhibition. This causes decreased T-cell activation and inflammatory cytokine release. Tacgraf 1mg Capsule 10 ‘S also inhibits the release of preformed mediators from skin mast cells and basophils. A decrease in T-cells aids in allogeneic transplantation. Tacgraf 1mg Capsule 10 ‘S works by weakening your body’s immune system to help your body accept the new organ as if it were your own.

How To Use Tacgraf 1mg Capsule 10 ‘S

Tacgraf 1mg Capsule 10 ‘S comes in capsule form. Take the Tacgraf 1mg Capsule 10 ‘S by mouth with or without food. It is usually taken every 12 hours or as directed by your doctor. If you experience nausea or upset stomach you may take the Tacgraf 1mg Capsule 10 ‘S with food. Take the Tacgraf 1mg Capsule 10 ‘S without crushing and swallow the whole capsule. 

Dosage

Dosage is based on your weight, medical condition, blood work, and response to therapy.

Primary Immunosuppression in liver and kidney allograft recipients.

Adults

Oral Liver Transplantation

  • The recommended dosage is 0.1-0.2 mg/kg/day in 2 divided doses. The regime is usually started approximately 6 hours after completion of transplant surgery. 

Kidney Transplantation

  • The recommended dosage is 0.15-0.3 mg/kg/day in 2 divided doses. The regime is usually started within 24 hours of transplant surgery completion. 

Heart Transplantation

  • With or without antibody induction: Starting within 5 days of transplantation but no earlier than 6 hrs. after transplantation. The recommended dosage is 75 mcg/kg daily in 2 divided doses. Oral therapy should start 8-12hrs. after IV infusion discontinued

Children

Oral Liver Transplantation

  • The recommended dosage in children is 0.3 mg/kg/day in 2 divided doses. 

Pregnancy and Lactation

If you are pregnant, think you may be pregnant, or are planning to have a baby, ask your doctor or pharmacist for advice before using Tacgraf 1mg Capsule 10 ‘S. 

Tacgraf 1mg Capsule 10 ‘S is contraindicated in lactation. 

Contraindications

Inform your doctor if you have a history of any of the following before using Tacgraf 1mg Capsule 10 ‘S:

  • Allergic to active ingredients of Tacgraf 1mg Capsule 10 ‘S.
  • Plan to receive and live vaccines.
  • Have or have had liver and kidney problems.
  • If you are pregnant or are planning a pregnancy.
  • If you are breastfeeding.

Precautions

Inform your doctor about any of the following conditions before using Tacgraf 1mg Capsule 10 ‘S

  • Hypertension (high blood pressure)
  • Mineral imbalances such as hyperkalemia (high potassium levels)

Common side effects in kidney transplant patients

  • Infections
  • Tremors

How To Cope with Adverse Effects

Pain- Take an over-the-counter pain medication. If pain persists, consult your doctor. 

Fever- Patients are advised to ingest over-the-counter painkillers (such as Paracetamol), drink plenty of fluids, and rest for a couple of days till the symptoms pass.

Headaches- If you have headaches, then rest and drink plenty of fluids. Avoid the use of alcohol. Take a prescribed painkiller. Consult your doctor if the headaches last longer than a week and turn into severe headaches. 

Feeling sick (nausea or vomiting) or diarrhea – Avoid eating rich or spicy food. It might help to take medication after food. If you’re sick, try drinking small, frequent sips of water. For diarrhea take plenty of fluids such as ORS to combat the water loss. 

Feeling dizzy (lightheaded) or weak – If the medication makes you feel dizzy or weak, stop what you’re doing and sit or lie down until you feel better. Avoid getting up quickly or immediately from a sitting or lying down position to a standing posture. Do not drive or use tools or machinery in case of any of these effects.

Tremors- Propranolol is usually given to treat essential tremors. But the use of propranolol to treat tremors due to Tacgraf 1mg Capsule 10 ‘S is not beneficial. Tremors due to Tacgraf 1mg Capsule 10 ‘S usually go away after the body adjusts to the level of Tacgraf 1mg Capsule 10 ‘S that your body needs.
